DNA甲基化修饰作为一种重要的表观遗传修饰, 能通过影响染色质结构, DNA构象、稳定性以及与蛋白质相互作用方式等, 起到调控基因表达的作用.在正常的生理条件以及一些疾病发生过程中均起着重要作用.DNA 甲基化是一种常见的表观遗传修饰, 由DNA甲基转移酶(DNA methyl-transferase, Dnmt) 催化, 以S-腺苷甲硫氨酸(SAM) 作为甲基供体而发生反应.催化反应有3 类类型, 包括将腺嘌呤转变为N-甲基腺嘌呤、将胞嘧啶转变为N- 甲基胞嘧啶以及将胞嘧啶转变为C-甲基胞嘧啶.原核生物中这3种类型均存在, 但是在高等真核生物中只存在第3种类型, 即将胞嘧啶(C) 转变为5-甲基胞嘧啶(5mC)。
本套装提供3种DNA标准品(200bp):未修饰的DNA(含胞嘧啶),甲基化修饰的DNA(含5-mC),和羟甲基化的DNA(5-hmC)。
The DNA Methylation/Hydroxymethylation Standard Set is designed to be used as positive controls in DNA methylation experiments and various applications including MS/hMS-PCR and quantification of cytosine modifications in any species. It can also be used as substrates for studies involving 5-methylcytosine (5mC) and 5-hydroxymethylcytosine (5hmC) interacting proteins.
